Microscopy and Fluorescence In-Situ Hybridization mini-study of four contaminant-degrading enrichment cultures and Biofilm formation study of the KB-1 and T3L cultures using confocal microscopy

چکیده
Four environmentally-relevant and contaminant-degrading enrichment cultures (KB-1, ACT 3, T3L, and ORCH4) from the Edward’s Laboratory at the University of Toronto were studied via traditional microscopy, laser scanning confocal microscopy, and scanning electron microscopy. Also, general bacteria and archaea probes were tested for all these cultures to determine their suitability for continuous monitoring by Fluorescence In-situ Hybridization (FISH). For the culture ACT 3, a Dehalobacter specific probe (Dhb_442) was designed in-silico and experimentally tested in order to determine the morphology of the Dehalobacter species in this culture as well as in the sub-culture WL 1,2 DCA. Based on the preliminary results obtained, the relative abundance of Dehalobacter in ACT 3 and WL 1,2 DCA was 27 and 39%, respectively. Biofilm formation for the cultures KB-1 and T3L was also studied via laser scanning confocal microscopy. Biofilm was identified in both cultures; however, KB-1 produced larger and thicker biofilms in the time period studied.

Achromobacter Species Isolated from Cystic Fibrosis Patients Reveal Distinctly Different Biofilm Morphotypes
Achromobacter species have attracted attention as emerging pathogens in cystic fibrosis. The clinical significance of Achromobacter infection is not yet fully elucidated; however, their intrinsic resistance to antimicrobials and ability to form biofilms renders them capable of establishing long-term chronic infections. Still, many aspects of Achromobacter biofilm formation remain uncharacterize...
متن کاملIn situ visualization of newly synthesized proteins in environmental microbes using amino acid tagging and click chemistry
Here we describe the application of a new click chemistry method for fluorescent tracking of protein synthesis in individual microorganisms within environmental samples. This technique, termed bioorthogonal non-canonical amino acid tagging (BONCAT), is based on the in vivo incorporation of the non-canonical amino acid L-azidohomoalanine (AHA), a surrogate for l-methionine, followed by fluoresce...
متن کاملThe in vivo dynamics of Streptococcus spp., Actinomyces naeslundii, Fusobacterium nucleatum and Veillonella spp. in dental plaque biofilm as analysed by five-colour multiplex fluorescence in situ hybridization.
The formation and composition of dental plaque biofilm in vivo are important factors which influence the development of gingivitis, caries and periodontitis. Studying dental plaque biofilm in in vitro models can cause an oversimplification of the real conditions in the oral cavity. In this study, bovine enamel slabs were fixed in an individual acrylic appliance in situ to quantify dental plaque...
متن کاملImaging of endodontic biofilms by combined microscopy (FISH/cLSM - SEM).
Scanning electron microscopy is a useful imaging approach for the visualization of bacterial biofilms in their natural environments including their medical and dental habitats, because it allows for the exploration of large surfaces with excellent resolution of topographic features. Most biofilms in nature, however, are embedded in a thick layer of extracellular matrix that prevents a clear ide...
متن کاملOral cavity anaerobic pathogens in biofilm formation on voice prostheses.
BACKGROUND A polymerase chain reaction (PCR)-based method has been used to identify oral anaerobic pathogens in biofilms on voice prostheses. The purpose of the present study was to determine the location of those pathogens inside the biofilms. METHODS Biofilms of 15 voice prostheses were sampled and used to identify the oral pathogens. Fluorescence in situ hybridization was applied on smears...
